Isolation, characterization and investigation of differentiation potential of human periodontal ligament cells and dental follicle progenitor cells and their response to BMP-7 in vitro.
The aim of this study was to assess the factors, mechanisms and the differences between periodontal ligament (PDL) cells and denta l follicle (DF) progenitor cells towards the osteoblastic/cementoblastic differentiation and to investigate the effects of BMP-7 on developmental (DF) and mature tissue-derived (PDL) cells, respectively. Primary cell culture of PDL cells and DF progenitor cells was performed. Osteogenic differentiation was evaluated using von Kossa, Alizarin Red S and immuno–histo–chemistry staining of osteocalcin.
Gene expression pattern was evaluated via real-time PCR. A series of CD surface marks were tested using flow cytometry and fluorescence-activated cell-sorting analysis was performed. Real-time RT-PCR demonstrated similar gene expression pattern of PDL cells and DF progenitor cells: the expression of OPN and OCN significantly was elevated when incubated with osteogenic components, Runx2 was unaffected, and Osteorix was hardly expressed whether in basic medium or induction medium. In addition, BMP-7 induced osteoblast/cementoblast differentiation of PDLSCs and DF progenitor cells in a dose- and time-dependent manner, as reflected by enhanced Runx2 and (OCN) mRNA transcript expression. BMP-7 triggers PDL cells and DF progenitor cells to differentiate towards an osteoblast/cementoblast phenotype.

[Analysis on clinical and pathological characteristics of 66 patients with stage IV breast cancer].
OBJECTIVE
To explore the clinical and pathological characteristics of stage IV breast cancer and to analyze their relationship with the morbidity and prognosis.
METHODS
The records of 66 patients presenting from January 2008 to December 2014 with stage IV breast cancer were reviewed. All of the patients were women and the median age was 57.5 (31 to 80) years, accounted for 3.01% (66/2 189) among the breast cancer patients treated in the same period. Statistical methods were used to analyze the correlation between clinical and pathological characteristics such as T-stage, N-stage, immuno–histo–chemistry and the morbidity and prognosis of stage IV breast cancer. The influence of patients characteristics to metastasis were compared by χ(2) test. Kaplan-Meier curves were reported for overall survival (OS), and the Log-rank test was used to compare the difference in groups. Cox proportional models were fitted for multivariate analysis.
RESULTS
The median survival time of stage IV breast cancer was 56.0 months and the 5-year survival rate was 40%. To metastasis, the effects of age, subtypes, histological grade, hormone receptor (HR) and human epidermal growth factor receptor 2 (HER2)had no significant statistics differences. It was concluded that the expression of HER2 (P=0.003) and HR (P=0.001) as well as single metastasis (P=0.029) were the influencing factors of the survival by multivariate Cox regression analysis. Primary tumor R0 surgery group and no surgery group had no significant statistics differences of overall survival and the 5-year survival rate (P=0.102).
CONCLUSIONS
Clinical and pathological characteristics have no effect on metastasis. The expression of HER2 and HR as well as single metastasis play important roles in survival.
Cell lines of human oral squamous-cell carcinomas retaining their differentiated phenotype.
Two cell lines from head-and-neck squamous-cell carcinomas (SCC) have been established and characterized. Cell line R105 was derived from a xenografted SCC of the floor of the mouth and cell line T87/rc from a SCC of the epiglottis. Identification of individual cytokeratins 4, 5, 7, 8, 10, 13, 14, 18 and 19 led to the conclusion that both cell types had squamous characteristics and that keratinization occurred in xenografts. Ultrastructurally, junctional complexes were observed in both cell lines. Characteristic marker chromosomes were found and although both cell lines were derived from male patients, the Y chromosome was missing from all examined cells. The basic biological parameters of both cell lines were modal chromosome numbers of 59 (R105) and 60 (T87/rc), a doubling time of 60 (R105) and 45 hr (T87/rc) and a DNA index of 1.54 (R105) and 1.31 (T87/rc).
The tumorigenicity of the 2 cell lines was proved by the ability to form colonies on a plastic substratum, as well as in a soft agar assay. Furthermore, the cells could produce multi-cellular tumour spheroids, and formed tumour nodules after subcutaneous inoculation into nude mice. The R105 tumour cells appeared to be better differentiated than the T87/rc as observed by histology and immuno(histo)chemistry. Both cell lines appear to retain SCC differentiation after being xenografted into nude mice, cultured for more than 40 passages in vitro and thereafter again xenografted into nude mice.
Tissue engineering in urinary bladder: morphological and functional characterization.
BACKGROUND
In the last years, tissue engineering has attracted lots of researchers, in urology too. This is due to the possibility to use this technique in several pathologies’ therapies, which generally require reconstructive surgical solutions. Our work’s aim is to evaluate morphological and functional aspects of cultivated urothelial and detrusorial tissues, both in “monolayer growth” and on scaffolds, in order to understand the chance of using them in reconstructive surgery.
METHODS
Tissue cultures of detrusorial and urothelial cells have been obtained from animals (pigs, rabbits) and men. The urothelial nature of obtained cells has been demonstrated by traditional histological observation (Hematoxylin – Eosin), by immuno-fluorescence assay (specific for cyto-keratins antibodies), by immuno–histo–chemistry techniques (using specific cyto-keratins 7, 17, and 20 antibodies). Detrusorial tissue has been studied by using antibodies specific for alpha-actin.
RESULTS
Urothelial and smooth muscle cells, when isolated and expanded in vitro, keep the typical characteristics of original tissue, as showed by classical histological observation (H-E), immuno–histo–chemistry and immuno-fluorescence assays. These results were positive both in monolayer colonies and on scaffolds. In vitro results are encouraging and they demonstrate that it is possible to obtain in vitro vesical tissue that could have analogous characteristics to the original organ; even though clinical utilisation of this technique must be more investigated, both in vitro and in vivo.
